ANGIOTENSINA-(1-7): EFEITO NA MATURAÇÃO IN VITRO DE OÓCITOS OVINOS
Objetivou-se avaliar o efeito da adição de angiotensina-(1-7) ao meio de maturação in vitro de oócitos ovinos. Setenta e quatro oócitos de ovários ovinos abatidos em frigorífico foram maturados in vitro: I – controle (n=22), II – Ang-(1-7) a 1 uM (n=24), III – Ang-(1-7) a 1 uM+A-779 a 1uM (n=28) em microgotas contendo 100 µL de meio de maturação, acrescido de 10% de SFB, sob óleo mineral em incubadora com atmosfera de 5% de CO2 em ar, a 38,5ºC por um período de 18 horas. Após este período, os oócitos foram desnudados e observados a extrusão do primeiro corpúsculo polar. A análise estatística foi realizada pelo teste t a 5% de probabilidade. a adição de angiotensina-(1-7) a 1 µM, bem como a adição de angiotensina-(1-7) a 1 µM juntamente com o seu antagonista específico A-779 na mesma concentração, diminuem a taxa de maturação in vitro de oócitos ovinos.

Abstract:
Aimed to evaluate the effect of addition of angiotensin-(1-7) in the middle of in vitro maturation of sheep oocytes. Seventy-four oocytes of sheep ovaries slaughtered in fridg were matured in vitro: I - control (n=22), II - Ang-(1-7) at 1 uM (n=24), III - Ang-(1-7) at 1 uM+A-779 at 1uM (n=28) in microdroplets containing 100 µL of maturation medium plus 10% fetal bovine serum under mineral oil in the incubator atmosphere 5% CO2 in air at 38.5° C for a period of 18 hours. After this period, the oocytes were denuded and observed the extrusion of the first polar body. Statistical analysis was performed using the t test at 5% probability. The addition of angiotensin-(1-7) at 1 uM, as well as the addition of angiotensin-(1-7) at 1 uM together with its specific antagonist A-779 at the same concentration, decrease the in vitro maturation rate ovine oocytes.
