BIOCONTROLE DO ÓLEO ESSENCIAL DE ORIGANUM VULGARE FRENTE ÀS LARVAS DE Aedes aegypti (DIPTERA: CULICIDAE)

O Origanum vulgare (orégano), pertence à família Lamiaceae, uma planta aromática conhecida pelo seu uso na culinária e propriedades medicinais. Seu óleo essencial, obtido por hidrodestilação, foi testado em larvas do mosquito Aedes aegpty e no ensaio de inibição da enzima acetilcolinesterase (AChE), afim de elucidar se a toxicidade ocorre por esse mecanismo, uma vez que esta enzima desempenha um papel fundamental na função neuromuscular dos insetos.Os constituintes químicos do óleo essencial de O. vulgare (OEOV) foram idenficados por cromatografia gasosa acoplada à espectrômetro de massas (CG-EM), cujos majoritários são: Terpinen-4-ol (52,54%); Carvacrol (12,96%); g-terpineno (8,27%); Timol (6,82%) e o β-cariofileno (5,89%). O OEOV inibiu a AChE  em 28,34 e 73,19%,  nas concentrações de 0,0625 e 1 mg mL-1,com uma CI50 de 0,2029 mg mL-1. Na atividade biocida realizada com larvas do A. aegypti no 3º e 4º estágio larval o OEOV apresentou concentrações Letais (CL50) de 85,730 e 85,193 ppm em e CL90 143,524 e 138,170 ppm em 24 e 48 horas respectivamente. Dessa maneira, os resultados observados no presente trabalho demostraram que o OEOV é promissor agente biocida contra larvas do 3º e 4º estágio de desenvolvimento do A.aegypti, onde se pode sugerir que a inibição da enzima acetilcolinesterase pode atuar como um dos mecanismos para esses resultados, tornando-os favoráveis para continuidade e o aprimoramento de futuros testes nessa linha de pesquisa.

  • Abstract:

    The  oregano (Origanum vulgare)belongs to family Lamiaceae, an aromatic plant known for its use in cooking and medicinal properties. Its essential oil obtained by hydrodistillation was tested in larvae of the mosquito Aedes aegypti, and in inhibition of acetylcholinesterase (AChE), in order to elucidate whether the toxicity occurs through this mechanism, since this enzyme plays a fundamental role in neuromuscular function of insects.The chemical constituents of the essential oil of O. vulgare (OEOV) were identified by gas chromatography coupled to mass spectrometry (GC-MS), whose majority are: Terpinen-4-ol (52.54%); Carvacrol (12.96%); g-terpinene (8.27%); THYMOL (6.82%) and β-caryophyllene (5.89%). The OEOV inhibited AChE  in 28.34 and 73.19%, in concentrations of 0.0625 and 1 mg mL-1,with an IC50 of 0.2029 mg mL-1. The biocidal activity performed with larvae of A. aegypti in 3º and 4º larval stage the OEOV presented Lethal Concentration (LC50) of 85,730 and 85,193 ppm and (LC90) 143,524 e 138,170 ppm, in periods of 24 and 48 houras. In this way, the results observed in the present study demonstrated that the OEOV is promising  biocidal agent against larvae of the 3rd and 4th stage of development of A.aegypti, it suggests that inhibition of the enzyme acetylcholinesterase act as one of the mechanisms for these results, making them favorable for continuity and improvement of future tests in this line of research.
